miR-26a and its Target CKS2 Modulate Cell Growth and Tumorigenesis of Papillary Thyroid Carcinoma

Abstract: BackgroundWhile many studies have shown that levels of miR-26a are lower in papillary thyroid carcinoma (PTC), the role and mechanism of miR-26a in PTC are unclear.MethodWe used database searches to select potential mRNA targets of miR-26a. Anti-miR-26a, miR-26a mimic, siRNA for CKS2 and their effects on cell growth, cell-cycle distribution and colony formation were evaluated. We also evaluate the over-expressed miR-26a in TPC-1 cells in severe combined immune-deficient mice. We used luciferase reporter assays… Show more

“…MiR26 has been shown to function as a tumor suppressor miRNA by targeting multiple genes, including EZH2 in nasopharyngeal carcinoma (35), Burkitt lymphoma (36), and lung cancer (37), IL6 in hepatocellular carcinoma (38), and CKS2 in papillary thyroid carcinoma (39). In this study, we show that miR26a and miR26b also function as tumor suppressor miRNAs in lung cancer, as miR26a and miR26b were significantly downregulated in lung cancer specimens compared with the paired normal tissues and miR26a and miR26b mimics inhibited the proliferation, migration, and invasion of lung cancer cells.…”

“…For instance, microRNAs function throughout post-transcriptional gene silencing by modulating the translation of mRNAs into proteins (29) and there are recent reports on epigenetic regulation by noncoding RNAs (30). Indeed, Lv et al reported that miR26a targets and represses CKS2 expression in papillary thyroid carcinoma (31).…”

“…In certain studies, a variety of results indicated that CKS2 is a direct target of specific genes. (35) explored the miR-26a-regulated target gene(s), using three publicly-available miRNA target prediction tools: TargetScan, ncRNA and mirecords. CKS2 was selected as a potential target.…”
