Mitotic index in the subrenal capsule assay as an indicator of the chemosensitivity of ovarian cancer

Abstract: The subrenal capsule assay (SRCA) is used in clinical oncology to assess the sensitivity of individual malignant tumors to various anticancer agents and their combinations. Mitotic indices reflect cancer cell proliferation and have prognostic value in epithelial neoplasms, including ovarian carcinoma. We combined the two tests (SRCA, mitotic index) by evaluating the numbers of mitotic figures per square millimeter of neoplastic epithelium (M/V) in paraffin-embedded tumor samples after SRCA. The M/V index was c… Show more

“…The subrenal capsule assay was carried out to evaluate the sensitivity of different cell lines (MCF7-con, MCF7-SATB1, MCF7 ⁄ ADR-con, and MCF7 ⁄ ADR-shRNASATB1) to ADM as described previously. (14) Briefly, exponentially growing cells (1 · 10 7 ) were harvested, resuspended in 250 lL serum-free medium, and then mixed with 12.5 lL fibrinogen (40 mg ⁄ mL) and 12.5 lL thrombin (200 lg ⁄ mL) to prepare the cell-fibrinogen clot. Nude mice, 8-12 weeks old, were anesthetized with sodium pentobarbital at a concentration of 70 mg ⁄ kg.…”

“…18 Briefly, exponentially growing cells (1ϫ10 7 ) were harvested, resuspended in 225 l serum-free medium and then mixed with 12.5 l of fibrinogen (40 mg/ml) and 12.5 l thrombin (200 /ml) to prepare cell-fibrinogen clot. Nude mice, 8 to 12 weeks old, were anesthetized with sodium pentobarbital at a concentration of 70 mg/kg.…”
