The ionizing-radiation-resistant fractions of two soil bacterial communities were investigated by exposing an arid soil from the Sonoran Desert and a nonarid soil from a Louisiana forest to various doses of ionizing radiation using a 60 Co source. The numbers of surviving bacteria decreased as the dose of gamma radiation to which the soils were exposed increased. Bacterial isolates surviving doses of 30 kGy were recovered from the Sonoran Desert soil, while no isolates were recovered from the nonarid forest soil after exposure to doses greater than 13 kGy. The phylogenetic diversities of the surviving culturable bacteria were compared for the two soils using 16S rRNA gene sequence analysis. In addition to a bacterial population that was more resistant to higher doses of ionizing radiation, the diversity of the isolates was greater in the arid soil. The taxonomic diversity of the isolates recovered was found to decrease as the level of ionizing-radiation exposure increased. Bacterial isolates of the genera Deinococcus, Geodermatophilus, and Hymenobacter were still recovered from the arid soil after exposure to doses of 17 to 30 kGy. The recovery of large numbers of extremely ionizing-radiationresistant bacteria from an arid soil and not from a nonarid soil provides further ecological support for the hypothesis that the ionizing-radiation resistance phenotype is a consequence of the evolution of other DNA repair systems that protect cells against commonly encountered environmental stressors, such as desiccation. The diverse group of bacterial strains isolated from the arid soil sample included 60 Deinococcus strains, the characterization of which revealed nine novel species of this genus.Extreme ionizing-radiation resistance has been observed in several members of the domains Bacteria and Archaea. The Geodermatophilaceae are unique model systems to study the ability to thrive on or within stones and their proteogenomes (referring to the whole protein arsenal encoded by the genome) could provide important insight into their adaptation mechanisms. Here we report the detailed comparative genome analysis of Blastococcus saxobsidens (Bs), Modestobacter marinus (Mm) and Geodermatophilus obscurus (Go) isolated respectively from the interior and the surface of calcarenite stones and from desert sandy soils. The genome-scale analysis of Bs, Mm and Go illustrates how adaptation to these niches can be achieved through various strategies including 'molecular tinkering/ opportunism' as shown by the high proportion of lost, duplicated or horizontally transferred genes and ORFans. Using high-throughput discovery proteomics, the three proteomes under unstressed conditions were analyzed, highlighting the most abundant biomarkers and the main protein factors. Proteomic data corroborated previously demonstrated stone-related ecological distribution. For instance, these data showed starvation-inducible, biofilm-related and DNA-protection proteins as signatures of the microbes associated with the interior, surface and outside of stones, respectively.

The Antarctic represents a largely untapped source for isolation of new microorganisms with potential to produce bioactive natural products. Actinomycetes are of special interest among such microorganisms as they are known to produce a large number of natural products, many of which have clinical, pharmaceutical or agricultural applications. We isolated, characterized and classified actinomycetes from soil samples collected from different locations on Signy Island, South Orkney Islands, in the maritime Antarctic. A total of 95 putative actinomycete strains were isolated from eight soil samples using eight types of selective isolation media. The strains were dereplicated into 16 groups based on morphology and Amplified Ribosomal DNA Restriction Analysis (ARDRA) patterns. Analysis of 16S rRNA gene sequences of representatives from each group showed that streptomycetes were the dominant actinomycetes isolated from these soils; however, there were also several strains belonging to diverse and rare genera in the class Actinobacteria, including Demetria, Glaciibacter, Kocuria, Marmoricola, Nakamurella and Tsukamurella. In addition, screening for antibacterial activity and non-ribosomal peptide synthetase genes showed that many of the actinomycete strains have the potential to produce antibacterial compounds.
