Modification of the Template Capacity of Chick‐Oviduct Chromatin for Form‐B RNA Polymerase by Estradiol

Abstract: 1.Within 2 h of administration of estradiol in vivo to immature chickens, RNA synthesis by both form A and B RNA polymerases is stimulated in isolated oviduct nuclei.2. 24 h after hormone treatment this effect is enhanced, and is accompanied by four-and two-fold increases in the level of extractable form A and B enzymes in the nucleus.3. The endogenous RNA polymerase activity associated with nuclear chromatin is also stimulated by estradiol. 9001, of this activity, as shown by the use of the drugs a-amanitin a… Show more

“…24 g of the internal lining (consisting primarily of tubular gland cells) were scraped from the outer muscle layer and used to prepare nuclei as outlined previously [9]. Chromatin was then prepared by sequential washing of nuclei through solutions of NaCl/EDTA, Tris-HC1 and 1.7 M sucrose as described for the preparation of immature oviduct chromatin [9], except that 100 mM NaCl containing 8mM EDTA pH 8.0 was used for the initial washes. The translucent chromatin pellet was resuspended in 10 mM Tris-HCl pH 8, assayed for DNA and protein [14,15], stored overnight a t -20 "C then slowly thawed a t 4 "C prior to its use in enzyme assays.…”

“…b) Assays of bacterial RNA polymerase using chromatin as template contained samples of chromatin and enzyme (1-6 units) plus 90 mM (NH4),S0,. c) Assays of form B RNA polymerase using chromatin as template contained chromatin (25 pg DNA) and enzyme (up to 0.5 units) plus 160mM The incorporation of [3H]UTP into acid-insoluble product was determined as described [9]. Blank assays containing enzyme but not DNA (for set a), containing chromatin but not enzyme (sets b, c) or precipitated a t time 0 (set d) were subtracted.…”
