“…Donor-dependent precondition of the nasal tissue has to be considered to explain these differences in demands on the differentiation medium. Primary cell differentiation, especially using respiratory epithelium, often suffers from variable outcomes with high donor-to-donor variability 34,35 . Donor-dependent variation was also observed in ELISA evaluation resulting in rather high standard deviations.…”

“…Furthermore, it should be considered that the possibility of donor variability for MSCs remains. 36 Human capillaries have an average diameter between 5-10 µm depending on the tissue they are located in. In both of our tested hydrogels, mean diameters of the formed structures are in the same range for all supporting cell types despite BM MSCs For these, the diameter was much smaller and the structures have to be considered nonfunctional.…”

“…First, isolation of MSCs from adult sources involves invasive and often painful procedures with possible donor site morbidity (Hass, Kasper, Böhm, & Jacobs, 2011). Second, they have variable proliferation and differentiation capabilities, which are further reduced upon passaging in vitro (Russell, Lefavor, Durand, Glover, & Zubair, 2018). Third, the self-renewal and differentiation potential of MSCs is severely affected by donor age, genetics, and exposure to environmental stress (D'Ippolito, Schiller, Ricordi, Roos, & Howard, 1999;Malgieri, Kantzari, Patrizi, & Gambardella, 2010;Mattiucci, Maurizi, Leoni, & Poloni, 2018).…”
