Triclosan (TCS) is a broad-spectrum antimicrobial compound that is incorporated into numerous consumer products. TCS has been detected in aquatic ecosystems across the U.S., raising concern about its potential ecological effects. We conducted a field survey and an artificial stream experiment to assess effects of TCS on benthic bacterial communities. Field sampling indicated that TCS concentrations in stream sediments increased with degree of urbanization. There was significant correlation between sediment TCS concentration and the proportion of cultivable benthic bacteria that were resistant to TCS, demonstrating that the levels of TCS present in these streams was affecting the native communities. An artificial stream experiment confirmed that TCS exposure could trigger increases in TCS resistance within cultivable benthic bacteria, and pyrosequencing analysis indicated that TCS resulted in decreased benthic bacterial diversity and shifts in bacterial community composition. One notable change was a 6-fold increase in the relative abundance of cyanobacterial sequences and a dramatic die-off of algae within the artificial streams. Selection of cyanobacteria over algae could have significant implications for higher trophic levels within streams. Finally, there were no observed effects of TCS on bacterial abundance or respiration rates, suggesting that bacterial density and function were highly resilient to TCS exposure.

The quantitative structure property relationship (QSPR) for the photolysis half-life (t 1/2 ) of polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans (PCDD/Fs) on spruce (Picea abies (L.) Karst.) needle surfaces under sunlight irradiation was investigated. Molecular distance-edge vector (MDEV) index was used as the structural descriptor of PCDD/Fs. The quantitative relationship between the MDEV index and log t 1/2 was modeled by using multivariable linear regression (MLR) and artificial neural network (ANN) respectively. Leave-one-out cross validation and external validation were carried out to assess the prediction ability of the developed models. For the MLR method, the prediction root mean square relative error (RMSRE) of leave-one-out cross validation and external validation is 3.47 and 4.25 respectively. For the ANN method, the prediction RMSRE of leave-one-out cross validation and external validation is 2.68 and 3.52 respectively. It is demonstrated that there is a quantitative relationship between the MDEV index and log t 1/2 of PCDD/Fs. Both MLR and ANN are practical for modeling this relationship. The developed MLR model and ANN model can be used to predict the log t 1/2 of PCDD/Fs. Thus, the log t 1/2 of each PCDD/F congener was predicted by using the developed models.

Soils contaminated with polychlorodibenzo-p-dioxins (PCDDs), polychlorodibenzofurans (PCDFs), and dioxin-like (dl) polychlorinated biphenyls (PCBs), known as persistent organic pollutants (POPs), have garnered global attention because of their toxicity and persistence in the environment. The standard method for target analytes has been used; however, it is an obstacle in large-scale sample analysis due to the comprehensive sample preparation and high-cost instrumental analysis. Thus, analytical development of inexpensive methods with lower barriers to determine PCDDs/Fs and dl-PCBs in soil is needed. In this study, a one-step cleanup method was developed and validated by combining a multilayer silica gel column and Florisil micro-column followed by gas chromatography with triple quadrupole mass spectrometry (GC-QqQ-MS/MS). To optimize the separation and quantification of 17 PCDDs/Fs and 12 dl-PCBs in soils, the sample cleanup and instrumental conditions were investigated. For quantification method validation, spiking experiments were conducted to determine the linearity of the calibration, recovery, and method detection limit of PCDDs/Fs and dl-PCBs using isotopic dilution GC-QqQ-MS/MS. The applicability of the simultaneous determination of PCDDs/Fs and dl-PCBs was confirmed by the recovery of native target congeners and labeled surrogate congeners spiked into the quality-control and actual soil samples. The results were in good agreement with the requirements imposed by standard methods. The findings in this work demonstrated the high accessibility of the sample cleanup and analysis methods for the efficient determination of PCDDs/Fs and dl-PCBs in contaminated soils.
