Modulation of apical constriction by Wnt signaling is required for lung epithelial shape transition

Abstract: In lung development, the apically constricted columnar epithelium forms numerous buds during the pseudoglandular stage. Subsequently, these epithelial cells change shape into the flat or cuboidal pneumocytes that form the air sacs during the canalicular and saccular (canalicular-saccular) stages, yet the impact of cell shape on tissue morphogenesis remains unclear. Here, we show that the expression of Wnt components is decreased in the canalicularsaccular stages, and that genetically constitutive activation of… Show more

“…Surfactant protein C (SP‐C), a downstream target of Nkx2.1 and a marker of distal pulmonary epithelial cells, can be detected as early as E9.5 . At E14.5, during the pseudoglandular stage, columnar cells prevail in the distal parts of the lung . By E16.5, alveolar progenitor cells, club cells (formerly known as Clara cells), and ciliated cells are detected (Figure ) .…”

“…While there are many agent-based models of other branching systems (e.g. Fumoto et al (2016); ; Lindenmayer (1968); Merks & Koolwijk (2009) ;Schatten et al (2007)) to our knowledge, this is the first agent-based model of kidney morphogenesis. A significant advantage of our approach is that unlike spatially-averaged compartment-based methods (e.g.…”
