Modulation of bone marrow mesenchymal stem cell secretome by ECM-like hydrogels

Abstract: It has been demonstrated that bone marrow mesenchymal stem cell (BM-MSCs) transplantation has beneficial effects on several central nervous system (CNS) debilitating conditions. Growing evidence indicate that trophic factors secreted by these cells are the key mechanism by which they are acting. These cells are frequently used in combination with 3D artificial matrices, for instance hydrogels, in tissue engineering-based approaches. However, so far, no study has been reported on the influence of such matrices,… Show more

“…Human fibroblasts 28 and adipose derived stem cellspeptide-modified alginate gels. More recently, furan functionalisation of GG, followed by a Diels-Alder reaction with a synthetic variant of the GRDGS peptide was used to direct neural and mesenchymal stem cell differentiation with encouraging results 30,31 . In this paper, we covalently linked unmodified peptides to GG, and monitored the reaction efficiency by tyrosine radiolabelling.…”

“…In this sense, our group has modified GG with GRGDS fibronectin peptide, which resulted in the enhancement of cell proliferation and metabolic activity, as will be described in detail in the next section [130, 190]. …”

“…Four weeks after transplantation, increased cell survival as well as migration throughout the hydrogel was observed, accompanied by functional improvement of the animal, in comparison to animals that received just BM-MSCs or a vehicle control of PBS [240]. More recently, GG was also suggested for MSCs encapsulation [190]. The engraftment of BM-MSCs within a GG hydrogel modified with GRGDS fibronectin-derived peptide as previously described [130] increased cell proliferation and metabolic activity, when compared to unmodified hydrogels.…”

“…In most cases, GG products need to be sterilised prior to use in cell culture and TE. Various techniques are in use for this purpose including autoclaving [34][35][36] , ethylene oxide treatment 37 , microfiltration 38 , and antibiotic supplements 39 . Although no studies directly compare these methods with respect to their impacts on GG, equivalent investigations using other polysaccharides [40][41][42][43] and ECM materials 40,44,45 provide a good indication of the likely effects on GG.…”
