Molecular Characterization of Selected Local and Exotic Cattle Using RAPD Marker

Abstract: In order to develop specific genetic markers and determine the genetic diversity of Bangladeshi native cattle (Pabna, Red Chittagong) and exotic breeds (Sahiwal), randomly amplified polymorphic DNA (RAPD) analysis was performed using 12 primers. Genomic DNA was extracted from 20 cattle (local and exotic) blood samples and extracted DNA was observed by gel electrophoresis. Among the random primers three were matched and found to be polymorphic. Genetic relations between cattle’s were determined by RAPD polymorp… Show more

“…Five of twenty primers able to produce clearly visible bands were selected. The clearance of bands depends on the quality of DNA the primary sequence which has more than 60% of GC suitable for RAPD analysis (Khatun, Hossain, & Mahbubur Rahman, 2012). Bands readability is also important in genetic diversity analysis.…”

“…Polymorphism between individuals is caused by sequence differences in one or both primer binding sites and can be seen from the presence or absence of certain RAPD bands (Khatun at al., 2012). The RAPD method is simple but has low reproducibility, another disadvantage is that it requires a wide variety of primers such as for forensic testing.…”

“…Several molecular markers like RAPD, RFLP and microsatellites are being used for molecular characterization of animal genetic resources [10][11][12]. In the last decade, mitochondrial DNA (mtDNA) has emerged as a very powerful and reliable marker in animal population and evolutionary biology [13][14][15][16].…”
