Molecular cloning and nucleotide sequence of a cDNA clone coding for rat brain myelin proteolipid

Abstract: A cDNA library from rat brain was constructed in pBR322 and screened with a 14-mer mixed oligonucleotide probe based on residues 231-235 of bovine proteolipid (PLP). A positive clone was isolated: it contained a 1334-base-pair cDNA insert and was subjected to DNA sequence analysis. The cDNA encoded information for the 276 amino acids of rat PLP. Comparison with bovine PLP sequence showed a complete amino acid sequence homology except for 4 amino acid residues. Rat brain Myelin ProteolipidOligonucleotiak probe … Show more

“…1). Our predicted sequence is in very good agreement with the previously determined portions of rat PLP sequence and with other rat cDNA clones (11,27). It is of interest that, as in the rat, multiple PLP-specific mRNAs are found in the mouse.…”

“…The expression of these myelin proteins correlates closely with the formation of myelin in the developing rat brain, and mouse mutants that are defective in myelin formation show lowered levels of these myelin proteins (4,14,20,25 expression and the genetic deficiency in the shiverer mouse myelination mutant (12,34,44). Several groups have recently isolated cDNA clones for the other major myelin protein, PLP, and have studied PLP mRNAs in bovine and rat tissue (11,13,27, 29). The current studies were undertaken to isolate PLP cDNA clones for studies on the developmental regulation of PLP expression in mice.…”

“…PLP is remarkably well conserved among mouse (this report), rat (1,16), cow (2), and human (C.P., L.D.H., and R.A.L., unpublished work) at the amino acid and nucleotide level. The protein encoded by the mouse PLP cDNA (Fig.…”

“…The protein encoded by the mouse PLP cDNA (Fig. 1) is identical to the rat (16) and human (C.P., L.D.H., and R.A.L., unpublished work) PLP and displays only two conservative amino acid differences from the bovine sequence (2 (18) or dideoxy chain termination (19) methods. The most 5' 300 bases of phage A PLP2, which extends from position one to the secondary polyadenylylation site at 2288, were also sequenced.…”

“…This modification most commonly occurs on cysteines (see, for review, Refs. 39, 45, and 46), although it has also been found on serine (47), threonine (48), and lysine residues (49). The cysteine residues in the cytoplasmic domain of TGF-␣ are potential sites of this type of lipid modification of transmembrane TGF-␣.…”
