Molecular cloning, characterization and gene expression of an antioxidant enzyme catalase (MrCat) from Macrobrachium rosenbergii

Macrobrachium rosenbergii commonly called giant freshwater prawn is a widely farmed crustacean in freshwater. Similar to other aquatic organisms, their growth and well-being is influenced by various physical, chemical and biological factors. We discuss about the critical growth limiting factors as well as disease causing agents and the potential immune molecules of M. rosenbergii that are proved to involve in preventing and/or responding to those limiting factors.

This study evaluated the expression level of antioxidant enzymes Cu, Zn-superoxide dismutase (Cu, Zn-SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione S-transferase (GST) in hemocytes of Macrobrachium rosenbergii exposed to copper by real-time PCR (qRT-PCR). Results showed that the mRNA expression of Cu, Zn-SOD increased to reach a peak at 6 h, then recovered to its normal level at 48 h. CAT expression level was significantly increased at 12 h and reached a peak at 24 h, but recovered to its normal level later. GPx expression level was significantly increased at 6 h and reached the peak at 12 h. GST expression level was significantly induced from 12 to 24 h and then dropped to its normal level at 48 h. These results indicated that antioxidant enzymes were inducible, possibly for removing excessive reactive oxygen species to protect prawn from oxidative stress.

Mud crab from the genus Scylla is also known as mangrove crab, which has been well-accepted as a good source of protein.Recently, the antioxidant properties present in mud crabs have been reported to have a part in the protection of cells against free radicals. Meanwhile, numerous antimicrobial peptides from mud crabs have managed to be characterized through the display of antimicrobial activities against Gram-positive and Gram-negative bacteria. Hence, this paper is an effort to collect recent literatures on antioxidant and antimicrobial properties in every part of mud crabs which include muscle tissue, hemolymph, and crab shell. Moreover, the effort to understand the biological properties of mud crabs is important to enhance its production in aquaculture industry. Therefore, this review hoped to attract the attention of natural product researchers to focus on the potential therapeutic applications of mud crabs.
