Molecular cytogenetic characterization and SSR marker analysis of a leaf rust resistant wheat line carrying a 6G(6B) substitution from Triticum timopheevii (Zhuk.)

Abstract: A disease (powdery mildew, leaf rust) resistant line was selected from the progenies of a Triticum aestivum 9 Triticum timopheevii amphiploid produced at Martonvásár. This line was previously identified with C-banding as a 6G(6B) substitution. In order to detect the 6G chromosome in a wheat background, fluorescence in situ hybridization (FISH) and microsatellite marker analysis were used. Ten microsatellite markers of the 43 tested generated PCR products that were polymorphic between chromosomes 6B and 6G, and… Show more

“…2 b). Like its female parent, T. timococcum carries the same species-specific translocations previously reported by Rodríguez et al ( 2000 ) and Uhrin et al ( 2012 ). The short arm of chromosome 6A t has a long translocated segment from the short arm of chromosome 1G, while 1G has a short segment from 6A t (6A t S/1GS).…”

“…Chromosome discrimination was carried out using the previously published FISH patterns of T. timopheevii (Jiang and Gill 1994 ; Uhrin et al 2012 ) and T. monococcum (Megyeri et al 2012 ). Chromosomes 6G, 6A t , 1A m and 5A m showed very strong pTa71 signals, while the green coloured pSc119.2 probe gave strong signals on all the G genome chromosomes and weaker signals on two A t chromosomes (1A t and 5A t ).…”

“…FISH is a powerful technique for localizing highly repetitive DNA sequences, detecting specific sites in particular regions of individual chromosomes and discriminate genome constitutions [ 32 , 33 ]. Many substitution lines related to common wheat chromosomes and alien chromosomes have been identified using FISH or nulli-tetrasomic analysis, including 6 G (6 B ) [ 34 ], 1St( 1 D ) [ 28 ], 6 Js /6 B [ 13 ], 2 R a (2 D ) [ 35 ], 2 Ns (2 D ) [ 36 ], 2 V b (2 D ) [ 37 ], and 3 D (3 Ns ) [ 18 ]. In this study, the standard FISH karyotype patterns of wheat parent 7182 was painted for the first time using oligonucleotide probes Oligo-pTa535 and Oligo-pSc119.2, which was convenient to distinguish common wheat chromosomes in M11003-3-1-15-8.…”

“…ponticum, where there is a lack of molecular biology information, the transfer of wheat molecular markers to the species is a feasible method for genetic analysis (Dobrovolskaya et al 2009). Over the last two decades, SSR markers have been used extensively as a effective tool for the genetic analysis of alien chromosomes in various species, such as Thinopyrum ponticum (Hu et al 2011), Thinopyrum intermedium (Lin et al 2007), Triticum timopheevii (Uhrin et al 2012), Leymus mollis (Zhao et al 2013) and Secale cereale (Tang et al 2008).…”
