Molecular docking based screening of triterpenoids as potential G-quadruplex stabilizing ligands with anti-cancer activity

Abstract: Triterpenoids isolated from Ganoderma lucidum (GLTs) exhibit a broad spectrum of anti-cancer properties, including anti-proliferative, anti-metastatic and anti-angiogenic activities. Current research studies revealed the role by GLTs in inducing apoptosis and suppression of telomerase activity of cancer cells with much lower toxicity to healthy cells. Compounds selectively binding and stabilizing G-quadruplex structures could inhibit the telomerase or downregulate the oncogenes and may act as anti-cancer agent… Show more

“…The most reactive oxygen species (ROS) include superoxide anion (O), hydroxyl radical (OH-), hydrogen peroxide radical (ROO-). The nitrogen derived free radicals are nitric oxide anion (NO-) and peroxynitrite anion (ONOO-) 53 . Most of the free radicals once produced are neutralized by cellular antioxidant defenses (enzymes and nonenzymatic molecules).…”

“…GA‐A has been documented to hold promising anticancer properties, embodying antiproliferative, antimetastatic, and antiangiogenic capacities with much lower side toxicity . Studies of Yang et al have suggested that GA‐A exerts antitumor activity against human breast cancer cell through regulating mitochondrial apoptosis, expression of cell‐cycle‐associated factors and proapoptotic signaling pathway.…”

“…The function of the zinc finger 439 protein remains unknown, but it is suggested to be involved in the regulation of gene transcription. Moreover, lucidenic acids may act as potential quadruplex stabilizing ligands and promising inhibitors of Bcl-2 [ 74 , 75 ], which is a well-known apoptosis suppressor.…”
