Molecular forms in human serum of enzymes synthesizing DNA precursors and DNA

Abstract: Both thymidine kinase (TK) and DNA polymerase (DNAp) are present in measurable amounts in human serum. Even though the use of TK as a clinical marker is rapidly increasing there has been no attempt to characterize the serum TK in a wider extent, i.e.; with respect to Mw or other biochemical parameters. Therefore sera with high TK or DNAp activities derived from patients with cytomegalovirus (CMV) infection, B12-deficiency and leukaemia were fractionated by gel exclusion chromatography. The TK activity eluted a… Show more

“…Using thymidine or its analogue 5-iodo-2'-deoxyuridine as a substrate, TK activity in serum (STK) was established as a proliferation marker in 1980 (36). Previous results indicate that STK1 is a polymeric form of TK1, probably existing in complex with other serum proteins, and has a total molecular weight of approximately 700 kDa (37). The STK activity is useful for the estimation of tumor spread and prognosis in patients with leukemia (38), Hodgkin and non-Hodgkin's lymphoma (36) or preoperative patients with advanced ovarian cancer (39), but not for prognosis and monitoring the outcome of tumor therapy in solid tumors (22).…”

“…Hydrogen peroxide was introduced to stop the TK enzyme and eliminate DTE. DTE or other sulfhydryl reagents are necessary for optimum serum TK activity (38 ), but they interfere strongly with the peroxidase activity.…”

“…Furthermore, modifications of serum TK1, such as those described earlier, will most likely alter the reactivity with different TK1 antibodies. It is known that TK1 is found in serum in a stable multimeric form (38 ), probably originating from disrupted proliferating tumor cells. It is possible that several epitopes will be masked in this protein conformation and that determinations using agents that bind to the TK1 protein or peptides derived from TK1 may thus give information other than direct activity measurements (23,29 ).…”

“…The reaction is performed at 37 • C in a 0.1 M sodium acetate phosphorylation buffer supplemented with enzyme co-factors and stabilizing reagents [11,23]. ATP serves as a phosphate donor in the reaction.…”
