Molecular Resolution Imaging of Dextran Monolayers Immobilized on Silica by Atomic Force Microscopy

Abstract: A range of dextrans (5, 10, and 70 kDa) were covalently bound to smooth silica slides and porous (500 Å) silica particles with an external diameter of 25 µm. AFM analysis of the slides in air has shown the presence of discrete, clustered, and in a few regions overlayered dextran molecules, which exhibit increasing diameter and decreasing packing densities with increasing molecular weight. Analysis of 10 and 70 kDa dextran bound to silica particles has revealed surface features consistent with those of the dext… Show more

“…Successful reactions include N,N'-dicyclohexylcarbodiimide (DCC) coupling of pepsin, [91] 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC) coupling of DNA, [177] the immobilization of pnitrophenyl chloroformate activated dextrans, [178] reaction of compounds containing acidic protons with diazobenzilidenefunctionalized glass slides, [179] and a photoaffinity reaction. [180] Many research groups have employed a two-step approach, where the amine-functionalized SAM was derivatized with a linker molecule after self-assembly to increase the reactivity of the monolayer.…”

“…Furthermore, interaction of the sample with areas of the probe tip other than the apex may lead to ''tip self-imaging'' and prevent optimum profiling of the edges of molecular features. 40 The results indicate that AFM can observe the aggregates and single chains simultaneously, since AFM is a sensitive instrument and the desired concentration is extremely dilute. In view of the above result, the size and shape of the aggregates and individual molecules of hyperbranched polysaccharide in two solutions have been determined by using SEC-LLS combined with microscopy.…”
