Molecular Sexing of the White-Winged Guan (<b><i>Penelope albipennis</i></b>) and Other Wild Birds of the North of Peru

Abstract: The use of accurate and reliable techniques for sex determination of wild birds is of special importance in captive breeding programs, especially in birds with monogamous, aggressive behavior, with absence of copulation, and with a low hatching rate. Using PCR, we evaluated the relative efficacy of primers HPF/HPR and CHD1Wr/NP/CHD1Zr in the amplification of the chromo-helicase-DNA binding 1 (CHD1) gene for sex determination in Penelope albipennis and 8 other species of cracids, 4 species of falconids, 4 speci… Show more

“…The growing concern for animal welfare has placed the industrial practice of eliminating male chickens in laying strains as one of the greatest ethical problems in modern poultry [22]. In this context, the use of accurate and reliable techniques for sex determination of endangered fowls is of special importance in breeding programs, especially in captive ones displaying monogamous behavior, with problems or absence of copulation, and with a low hatching rate [23]. In view of this situation, numerous research works are being carried out to develop a technique of sexing of chickens during the first days of embryonic development, since it has not yet developed sensitivity, and hence, there is no suffering [24].…”

“…The PCR approach used for sexing gave slightly different results than expected from the original protocol by Díaz Casana et al . (2019). For P. castanea only, female bird samples showed a single band at approximately 400 bp in the agarose gel instead of 2 bands (Fig.…”

“…The sex of all other samples was determined using a molecular methodology ( N = 74) of Díaz Casana et al . (2019). As a basis of comparison for the PCR approach, 2 samples for which sexing based on morphology was certain were randomly chosen for both sexes in each species ( N = 8).…”
