Molecular Toolbox for Genetic Manipulation of the Stalked Budding Bacterium Hyphomonas neptunium

Abstract: The alphaproteobacterium Hyphomonas neptunium proliferates by a unique budding mechanism in which daughter cells emerge from the end of a stalk-like extension emanating from the mother cell body. Studies of this species so far have been hampered by the lack of a genetic system and of molecular tools allowing the regulated expression of target genes. Based on microarray analyses, this work identifies two H. neptunium promoters that are activated specifically by copper and zinc. Functional analyses show that the… Show more

“…This Cu-binding protein may play a dual protective role by removing the excess of Cu from the periplasm by sequestration and by delivering this metal to CusCBA, thus enhancing its export. 50, 51 Here, a Cu chaperone (A0A0P7DXF0) and an outer membrane channel protein TolC (Z5XTK0) were identified. The last one was up-regulated in biofilm Cu-treated cells (FC +2.2).…”

“…The strains, plasmids and oligonucleotides used in this study are detailed in Supporting Information Tables S2-S6. H. neptunium was transformed by conjugation as described previously (Jung et al, 2015). Chromosomal in-frame deletions were generated by double-homologous recombination, using sacB-containing suicide vectors and a two-step procedure based on sucrose counterselection (Thanbichler and Shapiro, 2006).…”

“…To achieve this goal, we first engineered a replicating plasmid harboring an inducible promoter suitable for H. baltica . We adapted the system developed for a tightly controlled heavy metal (copper) promoter inducible system in Hyphomonas neptunium, a marine Caulobacterale closely related to H. baltica (35). Similarly, we used the promoter for copper resistant protein operon copAB (P cu ) in H. baltica ) copA, hbal_0699, and copB, hbal_0698 ) (Fig.…”
