Monocyte chemoattractant protein 1 released from glycosaminoglycans mediates its profibrotic effects in systemic sclerosis via the release of interleukin‐4 from T cells

Abstract: Objective. Monocyte chemoattractant protein 1 (MCP-1; CCL2) has been implicated in the pathogenesis of fibrotic diseases and is up-regulated in patients with systemic sclerosis (SSc). The aim of the present study was to examine the mechanisms by which MCP-1 mediates its profibrotic effects in the setting of SSc.Methods. The expression of receptors for MCP-1 on dermal fibroblasts was analyzed by real-time polymerase chain reaction and fluorescence-activated cell sorting. The ability of extracellular matrix prot… Show more

“…Total RNA was isolated with the NucleoSpin RNA II extraction system according to the instructions of the manufacturer (Machery-Nagel, Düren, Germany) and reverse transcribed into complementary DNA (cDNA) with random hexamers as previously described (21,22). Gene expression was quantified by TaqMan or SYBR Green real-time PCR using the ABI Prism 7700 Sequence Detection System (PE Applied Biosystems, Rotkreuz, Switzerland).…”

“…Peripheral blood mononuclear cells were isolated from patients with SSc (n ϭ 10) and healthy volunteers (n ϭ 13) by the Ficoll density-gradient method, as previously described (24). All patients fulfilled the criteria for SSc as proposed by LeRoy and Medsger (25).…”

“…No patients were taking disease-modifying antirheumatic drugs, corticosteroids, nonsteroidal antiinflammatory drugs, statins, or anticoagulants. Fibroblasts were prepared as previously described (14,15). All patients and controls signed a consent form approved by the local institutional review boards.…”
