Monofunctional PtII Complexes Based on 8‐Aminoquinoline: Synthesis and Pharmacological Characterization

Abstract: Among the heterocyclic compounds, 8-aminoquinoline and its derivatives have become important candidates for the preparation of new antiproliferative metallo-drugs. Here, we reported the synthesis and cytotoxicity evaluation of a series of platinum complexes using 8-aminoquinoline and its chiral 5,6,7,8-tetrahydro-derivatives as chelating ligands. In the proposed complexes, a differently and opportunely alkylated imidazole was used to prepare the corresponding monofunctional [a]

“…Apart from the action of these complexes on cell DNA and other molecular targets, as mentioned earlier, these compounds are characterized by their selectivity against cancer cells. This is related to the transporters that are involved in the transport of these complexes into the cell—cationic platinum complexes are transported via organic cation transporters (OCTs) that are overexpressed in cancer cells, for example, colorectal cancer [ 31 ].…”

“…It is worthy to note that complexes 2-5 are known as starting materials for the synthesis of compounds with potential applications in OLED and pharmaceutical materials. [13][14][15] Therefore, a easy procedure to synthesize them is very meaningful. In comparison to the previous literature, [13][14][15] complexes 2-5 in this study were produced in milder conditions with higher yields (90-95 %).…”

“…Based on our experience in the use of the chiral 8-amino-5,6,7,8-tetrahydroquinoline scaffold and its derivatives as ligands in transition metal complexes, and considering their employment as sources of chirality in organometallic catalysts for asymmetric reduction or for C-B bond formation reactions [38][39][40], we decided to focus our attention on the synthesis of three amino monophosphine ligands: amino-phosphine (L1), -phosphinite (L2) [41,42] and -phosphite (L3). For the synthesis of amino monophosphines, we started from the chiral 5,6,7,8-tetrahydroquinolin-ol, the enantiomers of which were successfully separated through enzymatic DKR by Candida antarctica lipase B with high yield and purity [43].…”
