Repeat expansions in the C9orf72 gene cause amyotrophic lateral sclerosis and frontotemporal dementia characterized by dipeptide-repeat protein (DPR) inclusions. The toxicity associated with two of these DPRs, poly-GR and poly-PR, has been associated with nucleocytoplasmic transport. To investigate the causal role of poly-GR or poly-PR on active nucleocytoplasmic transport, we measured nuclear import and export in poly-GR or poly-PR expressing Hela cells, neuronal-like SH-SY5Y cells and iPSC-derived motor neurons. Our data strongly indicate that poly-GR and poly-PR do not directly impede active nucleocytoplasmic transport.

The nuclear pore complex (NPC) is the sole gateway to the nuclear interior, and its function is essential to all eukaryotic life. Controlling the functionality of NPCs is a tremendous challenge for cells. Firstly, NPCs are large structures, and their complex assembly does occasionally go awry. Secondly, once assembled, some components of the NPC persist for an extremely long time and, as a result, are susceptible to accumulate damage. Lastly, a significant proportion of the NPC is composed of intrinsically disordered proteins that are prone to aggregation. In this review, we summarize how the quality of NPCs is guarded in young cells and discuss the current knowledge on the fate of NPCs during normal aging in different tissues and organisms. We discuss the extent to which current data supports a hypothesis that NPCs are poorly maintained during aging of nondividing cells, while in dividing cells the main challenge is related to the assembly of new NPCs. Our survey of current knowledge points toward NPC quality control as an important node in aging of both dividing and nondividing cells. Here, the loss of protein homeostasis during aging is central and the NPC appears to both be impacted by, and to drive, this process.

Asam fosfat merupakan merupakan asam mineral anorganik yang memiliki rumus kimia H3PO4. Asam fosfat juga dikenal pula sebagai asam ortofosfat atau asam fosfat(V). Asam fosfat berwujud zat padat dengan T.D =280°C dan T.L = 44,1°C dan pada temperatur tinggi (t =1040 °C), mengalami disosiasi. Sumber yang pada umumnya dari asam fosfat adalah larutan air 85%.Kelarutan senyawa asam fosfat menyebabkan terjadinya interaksi suatu sistem biologis yang mengamati sifat interaksi dari masing-masing molekul merupakan suatu proses yang sangat kompleks dan akan sulit jika melalui percobaan. Penelitian ini bertujuan untuk menganalisis interaksi molekuler dan kinetik dari senyawa asam fosfat. Metode yang digunakan adalah dengan pemodelan menggunakan chem office serta melalui jurnal hasil penelitian mengenai asam fosfat. Parameter yang digunakan untuk transformasi ionic asam fosfat adalah konduktivitas hidrolik yang dilakukan sebagai parameter untuk mengukur permeabilitas tanah yang digunakan pada pembuatan pupuk dengan metode yaitu (1) metode tinggi air konstan/constan head method (2) metode tinggi air konstan di dalam tangki/constan head soil core/tank method (3) metode tinggi air terjun di dalam tangki/falling head soil core/tank method dan (4) metode aliran air dalam kondisi kesetimbangan/steady flow soil column method. Sifat-sifat termokimia dari senyawa asam fosfat dalam fase liquid ∆𝑓𝐻0=-3055 kJ /mol,𝑆0 = 95,54 J/molK, dan ∆𝐺0= -3015 kJ/molK. Interaksi atau transformasi suatu ion dapat dilihat dari pengaruh aspek sifat termodinamika senyawa ion (asam fosfat) dalam suatu larutan melalui perhitungan viskositas, kecepatan hanyut,dan mobilitas yang akan mempengaruhi tingkat konduktivitas senyawa asam fosfat. Asam fosfat memiliki kecepatan hayut 0,41508 𝑉/𝑐𝑚, konduktivitas listrik 9,2 Ms/cm untuk persen massa 0,5%, mobilitas relative ion H+ dan PO43- adalah 1,62 dan 8,40 serta tingkat viskositas 3.86 mPa·s.
