Moringa oleifera Leaves Extract Protects Titanium Dioxide Nanoparticles-Induced Nephrotoxicity via Nrf2/HO-1 Signaling and Amelioration of Oxidative Stress

Abstract: The efficacy of Moringa oleifera leaf extract (MO) in alleviating nephrotoxicity induced by titanium dioxide nanoparticles (TiO NPs) was studied. Rats were divided into four groups. Group I received distilled water. Group II received TiONPs. Group III received both TiONPs suspension beside MO. Group IV received MO only. Kidney KIM-1, NF-кB TNF-α, and HSP-70 expression were significantly upregulated while both Nrf2 and HO-1were significantly downregulated in TiONPs-treated rats. MO decreases expression of KIM-1… Show more

“…Our data showed that mRNA expression levels of these three antioxidant genes were increased in cells treated with ME, indicating that they are involved in the intracellular antioxidant mechanism of ME. These findings were in agreement with other studies showing that moringa extract could inhibit the production of pro-inflammatory cytokines and induce the production of antioxidant enzymes in rats [49,53]. In fact, these antioxidant effects were attributed to the rich content of flavonoids (e.g., myricetin, quercetin, and kaempferol) and phenolic acids (e.g., gallic acid and chlorogenic acid) in ME.…”

“…Cellular antioxidant mechanisms such as expression of antioxidant enzymes can minimize oxidative stress. Emerging evidence suggests that the anti-inflammatory effect of ME might be attributed to its antioxidant properties [48,49,50]. To further explore this association, the antioxidant effects of ME were examined.…”

“…Of these factors, Nrf2, a master transcription factor, is transferred to the nucleus after activation, where it binds to the upstream cis-regulated antioxidant response element (ARE) sequence located in the promoter regions of genes responsible for cell protection (David et al, 2017). Nrf2 subsequently activates a variety of cellular protective enzymes, such as catalase (CAT), superoxide dismutase (SOD), glutathione (GSH-Px), and heme oxygenase-1 (HO-1) (Abdou et al, 2019). There is mounting evidence that the Nrf2 signaling pathway may represent a potential target for fighting oxidative stress-related cardiovascular diseases due to its significant antioxidant properties.…”

“…A wide degree of safety of different MO extracts was reported, such as for example the median lethal dose (LD 50 ) of alcoholic extract of MO leaves up to 5 gm/ kg body weight (bw) [21]. in vivo human and animals as well as in vitro studies proved that various extracts of MO leaves possess different therapeutic effects, such as anti-inflammatory, hypolipidemic, antioxidant, anti-diabetic, and chemoprotective (hepatic, cardiac, renal, and nervous) activities [11,[22][23][24][25].…”
