Moringin, A Stable Isothiocyanate from Moringa oleifera, Activates the Somatosensory and Pain Receptor TRPA1 Channel In Vitro

Abstract: Moringa oleifera Lam. is a tropical plant widely used in traditional medicines and as a food supplement. It is characterized by the presence of glucosinolates and isothiocyanates; the stable isothiocyanate 4-[(α-l-rhamnosyloxy)benzyl]isothiocyanate (moringin) has been widely studied for its bioactivity as hypoglycemic, antimicrobial, anticancer and in particular for its involvement in nociception and neurogenic pain. Moringa extracts and pure moringin were submitted to in vitro assays with the somatosensory TR… Show more

“…Reduction of the vascular permeability was also observed in this study (Dos Santos et al, 2018). Another in vitro program proved that moringin is a potent agonist of the TRPA1 channel (Borgonovo et al, 2020). Thus, the application of these analogs would benefit TMD treatment in a relatively safe way with few side effects.…”

“…The phytochemicals that have been recognized in the leaves of Moringa oleifera can be grouped in three main chemical families: flavonoids, biophenols and their derivatives, and glucosinolates (GLs) and their derivatives [23,24]. In particular, glucomoringin, belonging to the glucosinolate group, possesses several biological properties, being effective against oxidation, viruses, fungi and bacteria [25].…”

“…Several Nrf2-interacting nutrients are direct TRPA1 activators [ 21 , 110 ]. These include: (i) allyl isothiocyanates (AITC: pungent components of mustard, horseradish, and wasabi [ 2 ]), (ii) cinnamaldehyde from cinnamon [ 70 ], (iii) allicin, an organosulfur compound from garlic [ 111 ], (iv) green tea polyphenols [ 112 , 113 ], and (v) 3 glucosinolates from Sisymbrium officinale (isopropylisothiocyanate and 2-buthylisothiocyanate) or Moringa oleifera (4-[(α-l-rhamnosyloxy) benzyl] isothiocyanate) [ 114 , 115 ]. Sulforaphane, an AITC and the most potent natural Nrf2 activator, does not appear to interact with TRPA1.…”
