Idesia polycarpa Maxim is a native dioecious tree from East Asia cultivated for its fruits and as an ornamental plant throughout temperate regions. Given the economic potential, comparative studies on cultivated genotypes are of current interest. This study aims to discover the dynamic changes and potential functions of endogenous hormones in I. polycarpa, as well as the differences in endogenous hormone contents in different growth stages among different I. polycarpa provenances. We used High-Performance Liquid Chromatography (HPLC) to measure and compare the levels of abscisic acid (ABA), indole-3-acetic acid (IAA), gibberellin A3 (GA3), and trans-Zeatin-riboside (tZR) in the leaves, flowers, and fruits of I. polycarpa from various provenances between April and October. Our findings indicated that changes in the ABA and GA3 content of plants from Jiyuan and Tokyo were minimal from April to October. However, the levels of these two hormones in Chengdu plants vary greatly at different stages of development. The peak of IAA content in the three plant materials occurred primarily during the early fruit stage and the fruit expansion stage. The concentration of tZR in the three plant materials varies greatly. Furthermore, we discovered that the contents of endogenous hormones in I. polycarpa leaves, flowers, and fruits from Chengdu provenances were slightly higher than those from Tokyo and Jiyuan provenances. The content of IAA was higher in male flowers than in female flowers, and the content of ABA, GA3, and tZR was higher in female flowers than in male flowers. According to the findings, the contents of these four endogenous hormones in I. polycarpa are primarily determined by the genetic characteristics of the trees and are less affected by cultivation conditions. The gender of I. polycarpa had a great influence on these four endogenous hormones. The findings of this study will provide a theoretical foundation and practical guidance for artificially regulating the flowering and fruiting of I. polycarpa.

Idesia polycarpa Maxim is an emerging oil plant species. Understanding its microecological characteristics and internal mechanisms can serve as a basis for field management and the screening and application of growth-promoting bacteria during the growth phase of young trees. This study used three-year-old young I. polycarpa to analyze the tree’s root morphology, soil, and leaf nutrient status variations from May to October. In addition, differences in the rhizosphere soil, leaf metabolites, and microorganisms were observed. The results showed that, from May to October, the total nitrogen (TN) in the soil significantly decreased, whereas the TN, total potassium (TK), and total phosphorus (TP) in the leaves differed (p < 0.05). The leaf-dominant bacteria changed from Pseudomonadota to Firmicutes phylum. In addition, the relative abundance of soil and leaf-dominant bacteria decreased. The study found that the soil and leaf differential metabolites were mainly sugars and phenolic acids. The soil bacterial community showed a significant correlation with soil pH, available potassium (AK), available phosphorus (AP), and TN (p < 0.05). Further, the soil fungal community was significantly correlated with pH and AK (p < 0.001). TP, pH, and TK were the main factors influencing the leaf bacterial community, while the leaf fungal community was significantly correlated with five factors, including pH, TC, and TN. The root morphology was also mainly affected by pH, Pedomicrobium sp., Talaromyces sp., Penicillium sp., and D-Mannitol 2.

Idesia polycarpa is a dioecious tree; in field surveys, there are rare sex conversions in I. polycarpa individuals with bisexual flowers. To identify the molecular mechanisms underlying sex conversion in this species, an integrative analysis of the proteome and transcriptome profiles of I. polycarpa male and bisexual flowers at key developmental stages was conducted in this study using isobaric tags for relative and absolute quantification and RNA-seq technology. A total of 15,003 proteins were identified; the differentially expressed proteins (DEPs) were enriched in metabolic pathways, biosynthesis of secondary metabolites, and flavonoid metabolism pathways in all comparison groups. A total of 290,442 unigenes were obtained; these were compared with seven databases, revealing 196,366 annotated unigenes. In general, the expression of proteins and genes tended to be positively correlated, with Spearman correlation coefficients in the ranges of 0.152–0.262 (all genes and all proteins) and 0.497–0.778 (DEPs and DEGs). The integrative analysis of DEPs and DEGs between male and bisexual flowers revealed that the most significantly enriched pathways were flavonoid pathways, metabolic pathways, and the biosynthesis of secondary metabolites. Finally, four co-expressed proteins and transcripts and one gene associated with the flavonoid biosynthesis pathway were screened out. The proteins identified were p-coumaroyl shikimate 3′-hydroxylase, and shikimate/quinate hydroxycinnamoyl transferase, and the gene was caffeoyl-CoA O-methyltransferase. The analysis has revealed key potential proteins and genes involved in sex conversion at the molecular level and has provided a basis for future investigations of artificial regulation of sex differentiation in I. polycarpa.
