Morphological studies of rat brain tumors induced by N-nitrosomethylurea

Abstract: ✓ Experimental glial tumors were induced by weekly intravenous injections of N-nitrosomethylurea (NNMU) in rats. The tumors included low- and high-grade gliomas of the astrocytic series and mixed gliomas. The histology of the tumors did not vary significantly with serial passage through tissue culture, subcutaneous implantation, or freezing. These neoplasms provide reliable animal models of brain tumors common to man.

“…9L/Ro, a subline of the 9L N-methylnitrosourea-induced rat brain tumour (Schmidek et al, 1971), was grown as monolayer cell cultures in BME containing 10% FCS . Plating efficiencies of this cell line were determined on exponentially growing cell cultures 2 days after seeding 2 x 106 cells into 75Cm2 culture flasks.…”

“…Studies on rat cadavers showed no statistical difference in placement measurements between the stereotactic and template methods. These results indicate that the template method for intra cerebrally implanting tumour cells in rats provides a precise, accurate and rapid procedure that maximizes reproducibility with a significant reduction in labour requirements, when compared with the conventional stereotactic methodology. Keywords 9L transplantable brain tumour; Fischer rats; template implantationj alternative The rat 9L gliosarcoma brain tumour model has been used extensively in studies of radiotherapy, chemotherapy and mechanisms of brain cancer for more than 20 years (Schmidek et al 1971, Swenberg et al 1972 …”

“…It carries a very poor prognosis with mean overall survival of [12][13][14][15][16][17] months. Despite testing a multitude of chemotherapy agents and strategies, very little progress has been made in extending the life expectancy of those diagnosed with a GBM, since the establishment of the Stupp protocol (a combination of radiotherapy and temozolomide) a decade ago [2,3].…”

“…The C6 glioma cell line was grown out from a glioma induced in an outbred Wistar rat strain by repeated rou ds of treat e t ith N'N'-methylnitrosourea [11,12]. Injection of the C6 cell line into newborn rats produced a tumour with pathology consistent with a glioblastoma multiforme including a diffuse invading border [13].…”
