Morphological transformation induced by silver nanoparticles in a Balb/c 3T3 A31-1-1 mouse cell model to evaluate in vitro carcinogenic potential

Abstract: Carcinogenesis is a complex process involved in genotoxic and non-genotoxic pathways. The carcinogenic potential of silver nanoparticles (AgNPs) has been predicted by examining their genotoxic effects using several in vitro and in vivo models. However, there is no little information regarding the non-genotoxic effects of AgNPs related to carcinogenesis. The in vitro cell transformation assay (CTA) provides specific and sensitive evidence for predicting the tumorigenic potential of a chemical, which cannot be o… Show more

“…This is an interesting fact, since other AgNPs showed cytotoxicity at much lower concentrations. The concentration resulting in 90% inhibition of 3T3 cell viability of uncoated AgNPs (>100 nm) was 9.59 μg/mL [ 54 ]. The green synthesized silver nanoparticles (136 ± 10.09 nm) were achieved using the aqueous extract of Origanum vulgare (Oregano).…”

“…Continuous treatment without metabolic activation for 1.5-2.0 cell cycle length is also recommended (OECD TG 487) [64]. In the studies reviewed, treatment usually lasted 24 h or 48 h (40-44) with a few exceptions that lasted 4 h or 72 h [72,76,85]. Almost all studies tested uncoated AgNPs, except for some that used PVP-, citrate-or branched polyetherimide (bPEI)-coated AgNPs [72,74,84].…”

“…In 2015, the European Union Reference Laboratory for Animal Test alternatives also published a paper to test chemicals for carcinogenicity using an In Vitro Syrian Hamster Embryocell Transformation Assay ( Drasler et al, 2017 ; Dickinson et al, 2019 ). Wunhak Choo et al assessed the in vitro carcinogenic potential of Ag NPs by Balb/c3T3 A31-1-1 mouse model ( Choo et al, 2017 ). Sighinolfi et al evaluated the carcinogenic potential of metal NPs by using the BALB/3T3 cell transformation assay ( Sighinolfi et al, 2016 ).…”
