Morphological variations of patchouli putative mutants

Abstract: World patchouly oil need is almost 90% fulfilled by Indonesia as the main producer, however, their narrow genetic variation is a major obstacle in plant breeding. Chemical mutation such as colchine is believed to be able to wider patchouly genetic variation The aim of this study was to increase patchouly genetic variation using chemical mutagen. The variety of Patchoulina 2 was treated by colchicine on callus stage. Callus irradiated was grown into regeneration medium until formed plantlet and ready to acclima… Show more

“…However, this phenotype is reversible. An additional subculture period is required to move to normal morphology, as reported in previous studies (Suhesti et al, 2021(Suhesti et al, , 2020. LC 50 represents callus sensitivity, and giving a colchicine concentration of 0.0275% can cause 50% callus death.…”

“…Currently, roots, seeds, anther, callus, and apical meristem of the plant are source explants used to induce polyploidy. Previous research results show that colchicine induction through callus patchouli is most effective at a concentration of 0.02-0.06% with 2 hours of immersion (Suhesti et al, 2020). In Tagates erecta soaking the entire plants from roots to apical shoots with cotton in colchicine concentration of 0.0005% for 6 hours is the most effective treatment (Sadhukhan et al, 2014).…”
