Mouse RanBPM is a partner gene to a germline specific RNA helicase, mouse vasa homolog protein

Abstract: Germ cell-specific ATP-dependent RNA helicase, the product of the mouse vasa homolog (Mvh), has been shown to play an essential role in the development of the male germ cell. In male Mvh knockout mice, premeiotic germ cells arrest at the zygotene stage. To investigate the role of MVH protein in the progression of meiosis, we searched for genes encoding partners that interact with MVH in testicular germ cells. Using the yeast two-hybrid system, we found that MVH interacts with mouse RanBPM, a Ran-GTP binding pr… Show more

“…Among these were a SNc/Tudor domain protein (spot 3), a polyadenylation-binding protein (PABP; spot 10), and several RNA helicases (spots 10,15,22,23,30,32,37). Homologs of these proteins have been shown to associate with MTs, either directly or indirectly.…”

“…Homologs of these proteins have been shown to associate with MTs, either directly or indirectly. For example, PABP co-purified with MTs from sea urchin embryos and HeLa cells (30,31), and a human RNA helicase (Vasa) interacts with a MT-binding protein that regulates MT nucleation (32). A protein similar to the SNc/Tudor domaincontaining protein has also been identified in cytoskeleton fractions from rice and pea (33,34).…”

“…For example, the mouse vasa homolog gene (Mvh; Ddx4 -Mouse Genome Informatics), encoding an ATPdependent RNA helicase, is crucial for the development of male germ cells (Shibata et al, 2004;Tanaka et al, 2000). However, in Mvh-null males, germ cell differentiation stops earlier than in RanBPM -/-mice, at the zygotene stage of prophase I, and Mvh -/-females are fertile.…”

“…Vasa/MVH, a germ cell-specific RNA helicase, interacts with RanBPM, which recruits RanGTP. The resulting complex may play a role in trapping and transferring of specific RNAs from the nucleus to the chromatoid body (21). However it was noted that the binding of DDX3 to CRM1 did not required its N-terminal NES (rather a C-terminal sequences between aa 260 and 517) and RanGTP (22) for HIV-1 Rev-PRE export.…”
