Mouse Skin Inflammation Induced by Multiple Topical Applications of 12-O-Tetradecanoylphorbol-13-Acetate

Abstract: It is well known that applications of a single dose of 12-O-tetradecanoylphorbol-13-acetate (TPA) to mouse ears induces an acute inflammatory reaction consisting of erythema, edema and polymorphonuclear leukocyte (PMN) infiltration. We report here that multiple topical applications of TPA to mouse ears produce a prolonged inflammatory reaction characterized by increases in ear weight, inflammatory cell infiltration and epidermal hyperplasia. TPA was applied 5 times over 10 days to mouse ears. Epidermal thickne… Show more

“…15) Briefly, 20 µL of TPA solution (2.0 µg/ear×6 times) or acetone (vehicle) was topically applied to the inner and outer surfaces of the right ear with a micropipette on alternate days. On days 7-10, the mice were treated on the inner and outer surfaces of the right ear with CA (0.8, 1.6, and 2.4 mg/ear), vehicle (2% DMSO-20% propylene glycol-78% acetone, negative control), or dexamethasone (0.05 mg/ear in acetone, positive control) twice a day.…”

“…Induction of inflammation with 12-O-tetradecanoylphorbol-13-acetate (TPA; Sigma-Aldrich) started at day 0 with application of 20 l of 0.01% TPA suspended in acetone (10 l in each side of the ear) and repeated every 2 days, as described previously (19,20). A control group treated with acetone was included in each experiment.…”

“…Após o quinto dia do experimento, o gelcreme teste foi aplicado (1 mg/orelha), por via tópica, durante 4 dias (2 vezes ao dia), sendo o edema avaliado diariamente [23]. Os animais foram distribuídos em dois grupos: grupo controle (gel-creme base) e o grupo teste (gel-creme teste).…”
