Mps1 Phosphorylates Borealin to Control Aurora B Activity and Chromosome Alignment

Abstract: Maintenance of chromosomal stability relies on coordination between various processes that are critical for proper chromosome segregation in mitosis. Here we show that monopolar spindle 1 (Mps1) kinase, which is essential for the mitotic checkpoint, also controls correction of improper chromosome attachments. We report that Borealin/DasraB, a member of the complex that regulates the Aurora B kinase, is directly phosphorylated by Mps1 on residues that are crucial for Aurora B activity and chromosome alignment. … Show more

“…Here, we characterize Nbl1 as an S. pombe Borealin-like protein based on its association with known CPC components, its colocalization with other CPC subunits during mitosis, and its sequence and proposed structural similarity to human Borealin. Consistent with previous data directly linking Borealin to Aurora B activation (Jelluma et al, 2008), we additionally demonstrate that Nbl1 is required for Ark1 activity. Furthermore, we show that the S. pombe CPC influences the process of cytokinesis, at least partially by controlling the localization of the fission yeast Cdc14-family phosphatase, Clp1.…”

“…The relatively small time window in which Mps1 operates likely explains why the effect of Mps1 on Aurora B centromere localization was not previously detected [10,15].…”

“…6 Although the precise contribution of MPS1 to SAC functions remains elusive, 7 accumulating evidence suggests that MPS1 facilitates SAC activation by promoting the recruitment of SAC components to unoccupied KTs. [8][9][10][11][12][13][14][15] MPS1 has also been implicated in the so-called 'error correction mechanism', 16 a poorly characterized process that resolves the erroneous KT-MT attachments that frequently form in the early steps of mitosis. 17,18 According to current models, AURKB, one component of the chromosomal passenger complex (reviewed by Ruchaud et al 19 ), catalyzes this process by destabilizing incorrect attachments, hence generating free KTs that activate the SAC.…”
