mRNA expression patterns of insulin‐like growth factor system components in human neuroendocrine tumours

Abstract: The described differences of the expression patterns of the IGF system components in neuroendocrine tumour subtypes suggest tumour type-dependent different pathways in tumour growth control by IGF system components.

“…Because many human neuroendocrine tumors demonstrate high expression of IGF-1, IGF-2 and IGF-1 receptor, 20,21 we selected IGF-1, which is known to activate the PI3K/Akt/mTOR signaling pathway, to evaluate the effects of rapamycin on growth factor-stimulated cell proliferation. Rapamycin significantly decreased IGF-1-induced proliferation of both STC-1 and GLUTag cells.…”

“…IGFBPs modulate the bioavailability of IGFs and their role in cancer development is complex and bimodal (Pollak et al 2004). The majority of SI-NETs expresses IGF1, IGFR1 and IGFBPs (Nilsson et al 1993, Wulbrand et al 2000. Media from primary SI-NETs cultures also contained IGF1 and incubation with IGF1 induced proliferation in BON1 cells and IGFR1-positive primary cultures (Nilsson et al 1993, von Wichert et al 2000.…”

“…Media from primary SI-NETs cultures also contained IGF1 and incubation with IGF1 induced proliferation in BON1 cells and IGFR1-positive primary cultures (Nilsson et al 1993, von Wichert et al 2000. Conversely, only 1 out of 9 SI-NET specimens had expression of IGF2 or IGF2R (Wulbrand et al 2000). As regulation of IGF signalling is complex and its function during SI-NET-associated fibrogenesis is unknown, further studies are needed to determine the role of the IGF system in MF (Pollak et al 2004, Feghali-Bostwick 2005, Nishizawa et al 2016.…”

“…The IGF-I autocrine loop seems to be important in growth control of neuroendocrine tumors (NETs), particularly in functionally active histotypes (22,36). Von Wichert et al (35) described the presence of an IGF-I autocrine loop in BON cells, a functioning human gastroenteropancreatic NET cell line.…”

“…It is unknown if there is a role for IGF-II in the modulation of NET cell proliferation. However, IGF-II and IGF-IR are expressed in ϳ30 and 70% of NETs, respectively (36).…”
