mRNA Poly (A) Tail, a 3′ Enhancer of Translational Initiation

Abstract: To evaluate the hypothesis that the 3' poly(A) tract of mRNA plays a role in translational initiation, we constructed derivatives of pSP65 which direct the in vitro synthesis of mRNAs with different poly(A) tail lengths and compared, in reticulocyte extracts, the relative efficiencies with which such mRNAs were translated, degraded, recruited into polysomes, and assembled into messenger ribonucleoproteins or intermediates in the translational initiation pathway. Relative to mRNAs which were polyadenylated, we … Show more

“…The poly(A) tract has been shown to play a role both in translation and RNA stability in many systems (for reviews see [10][11][12]. The longer the poly(A) tract of eukaryotic cytosolic RNAs, the greater the translational efficiency of the RNA (13)(14)(15)(16). “…5). These proteins are enriched for biological functions such as translation initiation (eIF5A, eIF4B), ribosome biogenesis (RIOK1), and poly(A) RNA binding, which is also related to translation 58, 59 . The CK2 site in RIOK1 (Ser21/22) was critical for RIOK to support proliferation/survival of A549 cells.…”

“…This was achieved after acquiring the added tPA protein sequence from UniProtKB by using accession ID: P00750 [ 54 ]. Furthermore, to ensure stability of the mRNA-based vaccine design, the addition of elements specific to eukaryotic mRNAs [ 55 , 56 ] was performed. This included the addition of a 5′ cap, poly (A) tail, and 5′ and 3′ untranslated regions.…”
