mTOR Complex 1 Regulates Lipin 1 Localization to Control the SREBP Pathway

Abstract: SUMMARY The nutrient- and growth factor-responsive kinase, mTOR Complex 1 (mTORC1) regulates many processes that control growth including protein synthesis, autophagy, and lipogenesis. Through unknown mechanisms, mTORC1 promotes the function of SREBP, a master regulator of lipo- and sterolgenic gene transcription. Here, we demonstrate that mTORC1 regulates SREBP by controlling the nuclear entry of lipin 1, a phosphatidic acid phosphatase. Dephosphorylated, nuclear, catalytically active lipin 1 promotes nuclear… Show more

“…In addition, mTORC1 was shown to modulate the actions of mature SREBPs by regulating the localization of lipin‐1. When mTORC1 is suppressed, lipin‐1 translocates to the nucleus and inhibits nuclear accumulation of SREBP1 and SREBP2 (Peterson et al, 2011). …”

“…For example, mTORC1 also controls growth by negatively regulating autophagy through the direct phosphorylation of Unc-51-like kinase (ULK1; discussed below) (Chan 2009;Ganley et al 2009;Hosokawa et al 2009;Jung et al 2009). In addition, mTORC1 directly phosphorylates the phosphatidate phosphatase Lipin1 (which regulates lipid metabolism through SREBP1) and the growth factor receptor-bound protein 10 (Grb10; part of a negative feedback loop targeting insulin receptor signaling) (Hsu et al 2011;Peterson et al 2011;Yu et al 2011). In fact, two recent proteomic studies identifying Grb10 as an mTORC1 substrate further suggest that the insulin-stimulated phosphorylation cascade is largely mTOR-dependent (Hsu et al 2011;Yu et al 2011).…”

“…Studies indicate that mTORC1, which is activated by growth factors and amino acids, is a key sensor allowing cells and tissues to adapt their metabolism in response to the nutritional state (5). In the liver, it controls the activation of various metabolic processes including lipogenesis (6) and ketogenesis (3). Recent observations indicate that mTORC1 regulates mitochondrial biogenesis and metabolism (7,8), but the underlying mechanisms remain to be determined.…”
