Mucosal inflammatory cytokine production by intestinal biopsies in patients with ulcerative colitis and Crohn's disease

Abstract: Chronic inflammation in inflammatory bowel disease (IBD; Crohn's disease and ulcerative colitis) may be attributed partly to increased secretion of inflammatory cytokines. The aim of this study was to investigate simultaneously the spontaneous release patterns of tumor necrosis factor-alpha (TNF-alpha), interleukin-1-beta (IL-1 beta), and interleukin-6 (IL-6) by organ cultures of inflamed mucosa from IBD patients. Organ cultures of involved IBD mucosa spontaneously produced increased amounts of TNF-alpha, IL-1… Show more

“…These cytokines have previously been shown to be increased in media derived from organ cultures of colon tissues from patients with active IBD (23)(24)(25). This provides an additional validation of the use of organ culture methods used here to study the effects of experimental bromelain treatment on cytokine secretion.…”

“…G-CSF has previously been noted to be increased in organ culture supernatants of colon tissues from patients with IBD (24). G-CSF was demonstrated to delay apoptosis of neutrophils, an activity that could lead to accumulation of neutrophils in mucosal tissues of IBD patients (25).…”

“…TNF␣ is one of the principal mediators of the inflammatory response in mammals, transducing differential signals that regulate cellular activation and proliferation, cytotoxicity, and apoptosis (1,2). In addition to its role in acute septic shock, TNF␣ has been implicated in the pathogenesis of a wide variety of inflammatory diseases (3)(4)(5) as well as in the progression of alcoholic liver disease (6 -9). Data from a number of animal studies, in combination with clinical studies, indicate that longterm alcohol consumption results in the development of fatty liver and steatohepatitis with progression to more severe liver damage including fibrosis, cirrhosis, and hepatocellular carcinoma (9).…”

“…TNFA is highly expressed in the colon of IBD patients [34], anti-TNF antibodies are commonly used in the treatment of IBD. Blocking downstream TNFA products in a mouse-model has demonstrated a reduction in tumor formation [35].…”
