Multidrug resistance gene expression response to cisplatin and 5FU treatment in hepatoma, prostate and colon cancer cells

Abstract: Variable expression of drug transporters at the plasma membrane of tumor cells contributed to drug resistance. The aim of the present study was to explore the expression levels of MRP2 and MRP3genes in Cisplatin and 5 fluorouracil (5FU) treated Hep3B, PC3 and HT29 cell lines. MTT assay was used to determine the cytotoxic effects of cisplatin and 5FU. The results from the MTT assay revealed that PC3 cell line is the most sensitive for cisplatin treatment presenting the lowest IC50 value (0.5 µg/ml) for 48h. 5FU… Show more

“…Under the investigated experimental conditions, the IC 50 value for CisPt was found at the concentrations of 50-100 μM (data not shown), according to the spread values ranging approximately from 20 to 130 μM found in the literature. 7,43,44 These divergences can be explained by considering that prostate cancer cells, and specifically the PC-3 line, generate high levels of ROS including H 2 O 2 and superoxide, in turn, and the ROS level increases with the aggressiveness of the cells. 44 The higher values of the IC 50 determined by the MTT assay should be considered in this context, where the conversion of the tetrazolium salt i.e.…”
