Multifunctional Nanoparticles for Combined Doxorubicin and Photothermal Treatments

Abstract: To facilitate combined doxorubicin and photothermal treatments, we developed doxorubicin-loaded poly(lactic-co-glycolic acid)-gold half-shell nanoparticles (DOX-loaded PLGA-Au H-S NPs) by depositing Au films on DOX-loaded PLGA NPs. As the PLGA NPs biodegraded, DOX was released, and heat was locally generated upon near-infrared (NIR) irradiation due to NIR resonance of DOX-loaded PLGA H-S NPs. Compared with chemotherapy or photothermal treatment alone, the combined treatment demonstrated a synergistic effect, r… Show more

“…In addition, the combination of chemotherapy-loaded nanoparticles and photo-hyperthermia therapy allows to deliver anticancer drugs and heat to the tumorigenic regions, demonstrating a clear synergistic effect compared with chemotherapy or hyperthermia alone. Thus, Park et al [478] developed doxorubicin-loaded PLGA-gold half-shell nanoparticles by coating biodegradable doxorubicin-loaded PLGA nanoparticles with a film of gold. The multifunctional nanoparticles were endocytosed and accumulated within human cervical cancer (HeLa) cells, where particles were degraded, releasing the doxorubicin, and the heat was generated with near-infrared irradiation (NIR) over the gold layer that is NIR resonant.…”

“…PLGA NPs promise to be a successful system for the targeted and controlled release of doxorubicin with decreased systemic toxicity, increased therapeutic efficiency and patient compliance. Furthermore, multifunctional PLGA NPs for combined doxorubicin and photo thermal treatments were studied by Park et al to deliver both drug and heat simultaneously to a selected tumorigenic section (Park et al, 2009a;Acharya and Sahoo, 2011). “…DOX has been found to be more efficient if attached to hydrophilic nanoparticles which are able to enter into the cell more profoundly than the cytotoxic agent alone, and therefore they can increase uptake of free anthracycline drug (Guo et al, 2008;Patil et al, 2008;Wang, 2007). Moreover, in order to overcome various problems concerning nonspecific DOX delivery, some targeted drug deliveries and combined therapies have been investigated (Brulé et al, 2011;Park et al, 2009). The significance of this drug can be measured by its widespread commercial use, involving different tradenames, both for the drug itself (for example, Adriamycin and Rubex are used for the hydrochloride salt, and the brand name of liposome-encapsulated doxorubicin is Doxil or Myocet).…”
