Edwardsiella ictaluri, the causative agent of enteric septicaemia of catfish (ESC), expresses long O polysaccharide (OPS) chains on its surface. The authors previously reported the construction of an isogenic Ed. ictaluri OPS mutant strain and demonstrated that this strain is avirulent in channel catfish. This paper reports the cloning of the Ed. ictaluri OPS biosynthesis gene cluster and identification of the mutated gene in the OPS-negative strain. The sequenced region contains eight complete ORFs and one incomplete ORF encoding LPS biosynthesis enzymes. The mutated gene (designated wbiT ) was similar to other bacterial galactose-4-epimerases. Glycosyl composition analysis indicated that wild-type Ed. ictaluri OPS contains higher amounts of galactose and N-acetylgalactosamine than the OPS mutant strain, which correlated well with predicted functions of the genes identified in the OPS biosynthesis cluster. The OPS mutant had a relatively small, but significant, decrease in its ability to survive in normal catfish serum compared to wild-type Ed. ictaluri, but it retained the ability to resist killing by catfish neutrophils.

This paper describes an evolutionary analysis of ISI elements based on nucleotide sequence data from six iso-insertion elements of ISJ (isoISIs) which are present in chromosomes and plasmids of Escherichia coli and Shigella species as repeated sequences. The sequence comparison, which permitted construction of a phenogram, showed that the iso-ISIs can be divided into three groups. One group consists of four elements with 1 % nucleotide sequence divergence. A second and third group each consists of one element, with 10 % and 46 % divergence, respectively, to the first group. Despite their divergence, amino acid sequences in the two ISI encoded genes, insA and insB, and the terminal inverted repeat sequences, insL and insR, were found to be highly conserved. The evolutionary distance per site in the six sequences suggests that the ISI element has diverged to a greater degree than bacterial genes of known nucleotide sequences have. We postulate that the existence of a group of well conserved iso-ISIs and of highly diverged iso-ISIs may be due to the transposition ability of the ISJ element, generating repetitive sequences in both bacterial chromosomes and plasmids which can then independently diverge. We also discuss possible regulatory mechanisms of transposition mediated by ISI based on this analysis, including the influence of colon usage of insA and insB. The observed colon selection agrees well with those colons used by weakly expressed E. coli proteins.The insertion element ISI is one of a number of transposable DNA elements known in bacteria. SynopsisAdiabatic differential scanning microcalorimetry, which provides curves of the heat capacity vs temperature, was carried out for the DNA of plasmid pJL3-TB5 (5277 base pairs in length). The calorimetry curve shows nine peaks ranging from 81 to 96°C in 1 X SSC buffer at a heating rate of 0.25'C, due to the stepwise helix-coil transition of the DNA along the molecular chain. The theoretical melting curve, which can be constructed by calculation from the entire nucleotide sequence of the plasmid DNA by the helix-coil transition theory, is then compared with the calorimetry curve. The two curves resemble each other remarkably well, particularly when a parameter for the methylated adenine residues at GATC sites by Dam methylase is used appropriately. This allows us to assign each peak in the calorimetry curve to the melting of the respective regions of the plasmid DNA sequence. The local stability of the helix-coil transition along the DNA chain is closely related to the functional regions coded by pJL3-TB5, such as genes, transcriptional promoters, and particular sites generated by recombination of two different sequences in vivo and in vitro.
