Multiple induced mutagenesis for improvement of ethanol production by Kluyveromyces marxianus

Abstract: Kluyveromyces marxianus GX-15 was mutated multiple times by alternately treatment with UV irradiation and NTG for two cycles. Four mutant strains with improved ethanol yield were obtained. The maximum ethanol concentration, ethanol yield coefficient and theoretical ethanol yield of the best mutant strain, GX-UN120, was 69 g/l, 0.46 g/g and 91%, respectively, when fermenting 150 g glucose/l at 40°C. The corresponding values for GX-15 were 58 g/l, 0.39 g/g and 76%, respectively. GX-UN120 grew well in 11% (v/v) o… Show more

“…Induced mutants of Sc. stipitis for improved lignocellulose fermentation were generated in various studies (Watanabe et al ., 2010; Hughes et al ., 2011), whereas multiple rounds of mutagenesis boosted the ethanol production of Kluyveromyces marxianus (Pang et al ., 2010). …”

“…29 used UV irradiation and N-methyl-N′-nitro-N-nitrosoguanidine to mutate K. marxianus GX-15. They obtained a yield of 0.46 g ethanol/g glucose for the best mutant strain, K. marxianus GX-UN120, and this strain completely consumed all glucose at 60 h. In our work, the mutant strains 7-1 and 8-1 completely consumed all the glucose after 72 h and all the galacturonic acid after 168 h in comparison with the wild-type strain, which consumed 92% of the glucose and 96% of the galacturonic acid.…”

“…UV-C irradiation is a standard technique [27][28][29] for inducing mutations in yeast. It produces large numbers of random mutations broadly and uniformly over the whole genome to generate unique strains.…”

“…Among the natural yeasts that assimilate lactose, Kluyveromyces marxianus has been studied for ethanol production from cheese whey (Silveira et al 2005). Beyond hydrolysing lactose, K. marxianus ferments at shorter doubling times and higher temperatures than Kluyveromyces lactis (Lane et al 2011;Pang et al 2010). It is not clear why certain yeasts of the Kluyveromyces species are more fermentative than others (Lane and Morrissey 2010).…”
