Multiple roles for a novel RND‐type efflux system in Acinetobacter baumannii AB5075

Abstract: Colony opacity phase variation in Acinetobacter baumannii strain AB5075 is regulated by a reversible high‐frequency switch. Transposon mutagenesis was used to generate mutations that decreased the opaque to translucent switch and a gene encoding a predicted periplasmic membrane fusion component of a resistance–nodulation–cell division (RND)‐type efflux system was isolated. This gene was designated arpA and immediately downstream was a gene designated arpB that encodes a predicted membrane transporter of RND‐ty… Show more

“…In A. baumannii, some members of the RND superfamily have been implicated in the pathogenicity of the bacterium. For example, in A. baumannii strain AB5075, the attenuated virulence of two mutants (arpA and arpB) lacking components of an RND system was demonstrated in the G. mellonella model (22). Furthermore, inactivation of the gene A1S_0114, involved in the synthesis of a lipopeptide, reduces the motility and virulence of A. baumannii.…”

“…RND efflux systems, a category of bacterial efflux pumps, play a prominent role in both intrinsic and acquired multidrug resistance identified in Gram-negative bacteria and catalyze the active efflux of a wide variety of antibacterial substrates including antibiotics and chemotherapeutic agents [65,66]. RND efflux systems are normally composed of three proteins: an inner membrane transporter, an outer membrane protein that functions as a pore, and a periplasmic protein that interacts with both the inner and outer membrane proteins to provide a conduit for the extrusion of small molecules [67].…”

“…In particular, several regulatory genes, including RND-type efflux system, have been reported to be involved in the resistance and virulence of A . baumannii [ 13 ]. For example, the OmpA protein is involved in biofilm formation and virulence [ 14 ], it also can increase resistance to chloramphenicol, aztreonam, and nalidixic acid [ 15 ].…”
