Multiplexed suspension array platform for high-throughput protein assays

Abstract: A multiplexed suspension array platform, based on SU8 disks patterned with machine-readable binary identification codes is presented. Multiple probe molecules, each attached to individual disks with different unique codes, provide multiplexed detection of targets in a small sample volume. The experimental system consists of a microfluidic chamber for arraying the particles in a manner suitable for high throughput imaging using a simple fluorescent microscope, together with custom software for automated code re… Show more

“…2). 30 A sufficient number of accurate barcodes are critical to fulfill the high density analysis, and various encoding schemes have been used to this end, including spectrometric encoding, 12,15,18,30,31 graphical encoding, 19,[32][33][34][35] chemical encoding, [36][37][38][39][40] electronic encoding, 41,42 physical encoding, 43,44 and magnetic encoding. 45,46 Furthermore, the combination of different encoding techniques can produce an even larger amount of barcodes.…”

“…As for the majority of bead-based assays, reading the code and quantifying the target reporter signal on hydrogel particles can often rely on optical detection methods (for general reviews on bead-based array decoding see [83, 112, 116, 117]). Two approaches have been reported for signal acquisition: imaging methods [33, 34, 37, 38] and flow cytometry-like scanning techniques [44, 69].…”

“…Furthermore, the noncovalent attachment of the cholesteryl oligonucleotide has also been reported. 27 In addition, SU-8-encoded microparticles are chemically modied to perform multiplexed assays 16,28 or to be used for solid-phase synthesis. 24,25,29,30 Recently, a swelling-deswelling method for the surface modication of the 2D and 3D patterns of SU-8 was used, and demonstrated oligonucleotide attachment.…”
