Murine Gammaherpesvirus 68 Pathogenesis Is Independent of Caspase-1 and Caspase-11 in Mice and Impairs Interleukin-1β Production upon Extrinsic Stimulation in Culture

Abstract: Gammaherpesviruses establish lifelong infections that are associated with the development of cancer. These viruses subvert many aspects of the innate and adaptive immune response of the host. The inflammasome, a macromolecular protein complex that controls inflammatory responses to intracellular danger signals generated by pathogens, is both activated and subverted during human gammaherpesvirus infection in culture. The impact of the inflammasome response on gammaherpesvirus replication and latency in vivo is … Show more

“…Caspase-1 further cleaves proinflammatory cytokines interleukin-1b (IL1b) and interleukin-18 (IL18; ref. 17). Recent studies have revealed the roles of pyroptosis in several types of cancers, including colon cancer, gastric cancer, lung cancer, breast cancer, liver cancer, and glioma.…”

“…Little is known about the function of caspases in the replication cycles of other gammaherpesviruses or about the proviral roles of caspases in alpha-and betaherpesviruses. The inflammatory caspases caspase-1 and -11 are dispensable for the replication of murine gammaherpesvirus 68 (MHV68) and for MHV68 pathogenesis in vivo (55). Conversely, both human and mouse cytomegalovirus (CMV) activate caspase-1 through the DNA sensor absent in melanoma 2 (AIM2) (56)(57)(58).…”
