Mutations Altering the Mitochondrial-Cytoplasmic Distribution of Mod5p Implicate the Actin Cytoskeleton and mRNA 3′ Ends and/or Protein Synthesis in Mitochondrial Delivery

Abstract: ). A mutant allele of MOD5 encoding a protein (Mod5p-I,KR6) located predominantly in mitochondria was constructed. Mutants defective in delivering Mod5p-I,KR6 to mitochondria were sought by selecting cells with increased cytosolic activity of this protein. Twenty-five mutants defining four complementation groups, mdp1, mdp2, mdp3, and mdp4, were found. They are unable to respire at 34؇C or to grow on glucose medium at 38؇C. Cell fractionation studies showed that mdp1, mdp2, and mdp3 mutants have an altered mit… Show more

“…The control strain carrying an empty vector was not able to grow at 37°C. This is in line with a previous report demonstrating, suppression of another RSP5 mutation (mdp1-1) by overexpression of ubiquitin [17]. The defect of the rsp5-1 strain is caused by a single amino acid exchange in the HECT-domain of Rsp5 (L733S) [18].…”

“…Since ubiquitin chain formation is necessary for Vid vesicle function, but is dispensable for Vid24p binding to Vid vesicles, these results are consistent with the hypothesis that Vid vesicle formation is regulated by ubiquitin conjugation and ubiquitin chain formation. Thus, our work complements previous studies in which ubiquitin conjugation is important for peroxisome biogenesis (24), mitochondrial inheritance (25), mitochondrial targeting (26), and receptor-mediated endocytosis (27)(28)(29)(30)(31).…”

“…For example, the ubiquitin protein ligase Rsp5p is essential for mitochondrial inheritance and mitochondrial import (25,26). Rsp5p is also involved in receptor-mediated internalization of Ste2p, Ste3p, and other cell surface proteins (31).…”

“…Members of this highly-conserved family bind to OVCA1, a candidate breast and ovarian suppressor gene (Salicioni et al, 2000). Clone 26189, is 51% homologous to the human microsomal dipeptidase precursor (MDP1) which is identical to the ubiquitin -protein ligase gene (RSP5) (Zoladek et al, 1997). MDPI, a DNA-binding protein, inhibits the rapid growth of E-Coli and mycobacterium smegmatis, supporting the idea that it may function as a tumor suppressor gene (Matsumoto et al, 2000).…”
