Myocardin Overexpression Is Sufficient for Promoting the Development of a Mature Smooth Muscle Cell-Like Phenotype from Human Embryonic Stem Cells

Abstract: BackgroundMyocardin is thought to have a key role in smooth muscle cell (SMC) development by acting on CArG-dependent genes. However, it is unclear whether myocardin-induced SMC maturation and increases in agonist-induced calcium signalling are also associated with increases in the expression of non-CArG-dependent SMC-specific genes. Moreover, it is unknown whether myocardin promotes SMC development from human embryonic stem cells.Methodology/PrincipalFindings The effects of adenoviral-mediated myocardin overe… Show more

“…Indeed, Myocd alone can induce the mature SM marker SMMHC expression (Fig. 3, A and B), consistent with previous reports (24,36). Because Cdc7 played a very important role in regulating mature SM marker gene expression, and Cdc7 was induced much earlier than Myocd by TGF-␤ (20), we sought to determine whether Cdc7 induces mature SM marker expression through Myocd.…”

“…In subsequent work the same group determined that MYOCD is a master regulator of smooth muscle gene expression and is capable of activating smooth muscle expression in various non-muscle cell types by associating with SRF 9 . In a more recent study it was reported that forced MYOCD expression in human ESC derived-embryoid bodies only selectively regulated CArG-dependent smooth muscle genes and allowed an increase in the number of SMC-like cells derived following one month of differentiation 40 . Altogether, these reports align with our findings that transient over-expression of MYOCD can convert EPC to SMC.…”

“…In contrast, myocardin overexpression promotes VSMC differentiation and increases the expression of other VSMC marker proteins (38). Factors that inhibit myocardin expression are associated with dedifferentiation (35).…”
