Nanoparticles decorated with folate based on a site-selective αCD-rotaxanated PEG-b-PCL copolymer for targeted cancer therapy

Abstract: NPs fabricated from a mixture of PEG-b-PCL and selectively rotaxanated Fol-PEG(αCD)-PCL showed internalisation in KB cells through an active targeting mechanism.

“…By refining the reaction domains, formulations with increased binding affinities can be produced, optimizing ligand orientation for maximal interaction and enhancing the effectiveness of the covalent conjugation. [87,88] Nonetheless, strong covalent binding faces limitations arising from the control of environmental conditions during functionalization, such as pH [89] as well as non-specific interactions between living systems and the nanoparticles. [90] The complex nature of wet-chemical covalent conjugation approaches remains their major drawback.…”

“…For its low toxicity, low immunogenicity, and high affinity to folate receptors (FR) overexpressed in a variety of cancer cells, FA (folic acid) has been widely used as an excellent targeting agent to improve the delivery of anticancer drugs or gene products [ 48 , 49 ]. FA-modified PEG could increase the cell uptake of PR nanoparticles in KB cells [ 50 ]. Additionally, FA introduced to cationic PRs could achieve enhanced efficiency of gene delivery.…”

“…Hence, PCL copolymers have widely been used in basic and clinical investigations. For instance, PEG-PCL copolymer is a proper biomaterial that has steadily been utilized for therapeutic purposes [28][29][30] . As another example, a blended PCL with polypropylene or polyethylene and spun into fibers develop a notably efficient pigment dispersing aid 31 .…”
