Nanoplastic Exposure at Predicted Environmental Concentrations Induces Activation of Germline Ephrin Signal Associated with Toxicity Formation in the Caenorhabditis elegans Offspring

Abstract: In nematode Caenorhabditis elegans, exposure to polystyrene nanoparticles (PS-NPs) at predicted environmental concentrations can cause induction of transgenerational toxicity. However, the underlying mechanisms for toxicity formation of PS-NP in the offspring remain largely unknown. In this study, based on high-throughput sequencing, Ephrin ligand EFN-3 was identified as a target of KSR-1/2 (two kinase suppressors of Ras) in the germline during the control of transgenerational PS-NP toxicity. At parental gener… Show more

“…The qRT-PCR was carried out using SYBR Green Master Mix in an ABI 7500 real-time PCR system. The method of comparative CT (ΔΔ CT) was used to analyze alterations in transcriptional expressions of examined genes after the normalization with expression of reference gene ( tba-1 for C. elegans and pvdQ for P. aeruginosa ) ( Yang D. et al, 2021 ; Zhao Y. et al, 2022 ). Three replicates were performed.…”

“…Studies using laboratory-produced nanoplastics have already tested the ecotoxicity of nanoplastic for soil organisms such as C. elegans , identifying energy budget impairment and decreases in reproductive fitness [ 176 ] and testing C. elegans nanoplastic laboratory-produced exposure at predicted environmental concentrations [ 177 , 178 ].…”
