CD-nanosponges were prepared by crosslinking B-CD with diphenylcarbonate (DPC) using ultrasound assisted technique. 5-FU was incorporated with NS by freeze drying, and the phase solubility study, complexation efficiency (CE) entrapment efficiency were performed. Also, the particle morphology was studied using SEM and AFM. The in-vitro release of 5-FU from the prepared nanosponges was carried out in 0.1N HCl. 5-FU nanosponges particle size was in the nano size. The optimum formula showed a particle size of (405.46±30) nm, with a polydispersity index (PDI) (0.328±0.002) and a negative zeta potential (-18.75±1.8). Also the drug entrapment efficiency varied with the CD: DPC molar ratio from 15.6 % to 30%. The SEM and AFM showed crystalline and porous nature of the nanosponges. The in vitro drug release study of the selected formula 5-FUNS2 exhibited the fastest dissolution rate which is 56% in the first hr. Different molar ratios of (cyclodextrin to crosslinker) (CD: DPC) has a proficient effect on complexation efficiency (CE), apparent stability constant (Kst) and entrapment efficiency of 5-FU. 5-FUNS2 with (1:4) molar ratio showed the best result of CE, Kst and entrapment efficiency. 5-FUNS2 gave a higher release rate than the 5-FU-BCD inclusion complex and 5-FU solution. Surface morphology of the prepared nanosponges by SEM, AFM indicate that nanosized and highly porous nanosponges was obtained. The overall results suggest that cyclodextrin nanosponges could be a promising 5-FU delivery system utilizing the suitable formula.

Polymers constitute the most important group of excipients utilized in modern pharmaceutical technology, playing an essential role in the development of drug dosage forms. Synthetic, semisynthetic, and natural polymeric materials offer opportunities to overcome different formulative challenges and to design novel dosage forms for controlled release or for site-specific drug delivery. They are extensively used to design therapeutic systems, modify drug release, or mask unpleasant drug taste. Cellulose derivatives are characterized by different physicochemical properties, such as swellability, viscosity, biodegradability, pH dependency, or mucoadhesion, which determine their use in industry. One cellulose derivative with widespread application is ethylcellulose. Ethylcellulose is used in pharmaceutical technology as a coating agent, flavoring fixative, binder, filler, film-former, drug carrier, or stabilizer. The aim of this article is to provide a broad overview of ethylcellulose utilization for pharmaceutical purposes, with particular emphasis on its multidirectional role in the development of oral and topical drug dosage forms.

The contemporary development in the drug delivery system is a seamless process. Any delivery system targeting a molecule to a particular site is always been appreciated. This requirement can be fulfilled through a specialized drug delivery system that can easily target the drug at the site of action without compromising its efficacy and quality. Few new drugs typically cannot be effectually delivered by conventional dosage form. Hence, benefits from targeted, localized delivery of therapeutic agents are other driving forces for the current scenario. Nanosponge technology has been introduced to assist the release of drug in a controlled manner over time to reduce systemic toxicity and severe reactions. Nanosponges are tiny sponges with a size of about a virus (250 nm-1 μm), which consist of cavities that can be filled with a wide variety of hydrophilic as well as hydrophobic drugs, which can be further incorporated into a pharmaceutical dosage form such as oral, parenteral, topical or inhalation. The sighting of nanosponge has become a noteworthy step in overcoming certain problems such as drug toxicity, poor bioavailability, physiochemical instability , and patient non-acceptability. INTRODUCTION: Authentic targeted drug delivery systems have been a fantasy for always in the past and today as well. But it has been mainly unsatisfied by the difficult chemistry that is involved in the new development systems. Targeting drug delivery has long been problematic for medical investigators, i.e., how to get them to the right place in the body and how to control the release of the drug to prevent overdoses.
