Natural Compounds and Derivatives as Ser/Thr Protein Kinase Modulators and Inhibitors

Abstract: The need for new drugs is compelling, irrespective of the disease. Focusing on medical problems in the Western countries, heart disease and cancer are at the moment predominant illnesses. Owing to the fact that ~90% of all 21,000 cellular proteins in humans are regulated by phosphorylation/dephosphorylation it is not surprising that the enzymes catalysing these reactions (i.e., protein kinases and phosphatases, respectively) have attracted considerable attention in the recent past. Protein kinases are major te… Show more

“…Polyphenols have been known for their potential of targeting protein kinases via interacting with the ATP-binding sites ( Guerra and Issinger, 2019 ). Hispolon inhibited NPC cell metastasis by downregulating Akt signaling pathway ( Ho et al, 2017 ).…”

“…Phenolic compounds can inhibit the catalytic activity of protein kinases involved in MAP kinase and NF-κB pathways [141]. In addition, they can inhibit the catalytic activity of metabolic enzymes involved in prostaglandin synthesis [142,143].…”

“…At least 18 of these kinase inhibitors inhibit a number of kinases; that is, they are multi-target inhibitors [ 9 ]. Metabolites from plants are known to be a rich source of putative protein kinase inhibitors (e.g., flavonoid compounds that function as competitive inhibitors of ATP binding [ 13 ]).…”
