Natural Products as a Source for New Anti-Inflammatory and Analgesic Compounds through the Inhibition of Purinergic P2X Receptors

Abstract: Natural products have reemerged in traditional medicine as a potential source of new molecules or phytomedicines to help with health disorders. It has been established that members of the P2X subfamily, ATP-gated ion channels, are crucial to the inflammatory process and pain signalization. As such, several preclinical studies have demonstrated that P2X2R, P2X3R, P2X4R and P2X7R are promising pharmacological targets to control inflammatory and pain disorders. Several studies have indicated that natural products… Show more

“…192 Several natural products are shown to cause analgesia on inflammatory pain or neuropathic pain by inhibiting P2X 3 or P2X 7 mechanisms, although the selectivity of these compounds for specific P2X receptors remains unclear. Paeoniflorin ( 72 ) is a chief ingredient in the root of Paeonia lactiflora Pall, and it has been shown to be effective in relieving colorectal distension induced visceral pain and hyperalgesia in a rat model of IBS (neonatal maternal separation).…”

“…As a neurotransmitter, ATP plays an important role in regulating the transduction of the visceral pain signal by binding to P2X receptors [15,16], and ATP participates in the regulation of intestinal movement and gastrointestinal secretion. P2X receptors are widely distributed in the body, and they play an important role in the formation, transduction, and regulation of neuropathic pain, inflammatory pain, and visceral pain [17][18][19][20]. Many studies have shown that purinergic (P2X) receptors are involved in pain signal transmission, in which the P2X 3 and P2X 2/3 receptors are the main subtypes [21,22].…”

“…Of the seven subtypes of P2XR, the types that are most related to the progression or control of pain status are the P2X3R, the heteromericeP2X2/3R, P2X4R, and the P2X7R [78].…”
