Nef: “Necessary and Enforcing Factor” in HIV Infection

Abstract: The Human Immunodeficiency Virus -1 (HIV-1) Nef protein that was originally identified as a viral negative factor is a 27kDa myristoylated protein. However, this so called dispensable viral protein has emerged as one of the most important proteins for viral life cycle. Nef not only establishes the host cell environment suitable for viral replication and pathogenesis but also facilitates the progression of the infection into disease. Previous efforts have been focussed to explain how Nef down modulates host cel… Show more

“…The complex between CycK and CDK9 was also shown to function as a CTD kinase in vitro (17). Although Nef was earlier reported as a negative factor for viral replication (32), recent evidence has convincingly demonstrated Nef as an enhancer of viral replication (16,22,(33)(34)(35)(36). Nef performs most of its functions by interacting with cellular proteins.…”

“…Nef is an accessory protein encoded by the human (HIV-1 and HIV-2) 2 and simian immunodeficiency viruses and is an essential mediator of viral pathogenicity (1)(2)(3). Experimental deletion within the simian immunodeficiency virus nef gene reduces viral load, delays the onset of AIDS-like disease, and offers immune protection against challenge with pathogenic simian immunodeficiency virus in rhesus macaques (4,5).…”

“…The LTNP condition depends on a complex interplay between host and viral factors (1,3,19,36,47,52). Infection with HIV-1 variants with attenuated replicative capacity may contribute to the beneficial course of disease in a minor group of LTNP (7).…”
