Negative regulation of FcϵRI-mediated mast cell activation by a ubiquitin-protein ligase Cbl-b

Abstract: IntroductionMast cells play a central role in inflammatory and immediate allergic reactions. The aggregation of Fc⑀RI triggers the activation of Lyn protein-tyrosine kinase (PTK), resulting in the rapid tyrosine phosphorylation of its ␤ and ␥ subunits. 1 We isolated nonreceptor type PTK Syk. 2,3 Tyrosine phosphorylated ␥ subunit of Fc⑀RI then recruits and activates Syk, the essential PTK for antigen-induced Ca ϩϩ influx, degranulation, and cytokine production in mast cells. [3][4][5][6][7][8][9] Syk phosphoryl… Show more

“…Thus, certain drugs that enhance the activity of the Cbl protein family may provide new strategies for cancer therapy (19)(20)(21). Our previous studies found that up-regulation of Cbl-b inhibited Akt and ERK activation, and thus increased the sensitivity of different drugs in gastric cancer and leukemia cells (22,23).…”

“…RBL-2H3 cells overexpressing DN Cbl-b have been used in previous studies (27,30). It was observed that p-Akt expression is slightly increased in Cbl-b (DN) overexpressing cells.…”

“…Rat basophilic leukemia RBL-2H3 cells and cells overexpressing a dominant negative (DN) Cbl-b mutation (27) were maintained as monolayer cultures in Dulbecco's modified Eagle's medium (DMEM) containing 100 U/mL penicillin and 10% heat-inactivated FBS (27). The transfected cells were cultured in the presence of 0.4 mg/mL G418 (USA).…”

“…Some previous surveys showed that Cbl-b sequestered signalling molecules from lipid rafts, which resulted in ineffective lipid raft aggregation in mast cells [18,19].…”
