Neuron-derived Neurotrophic Factor Functions as a Novel Modulator That Enhances Endothelial Cell Function and Revascularization Processes

Abstract: Background: Tissue ischemia stimulates production of secreted factors that regulate angiogenesis. Results: Neuron-derived neurotrophic factor (NDNF) is up-regulated in endothelial cells in ischemic limbs of mice. NDNF stimulates endothelial cell function and promotes ischemia-induced revascularization through NOS-dependent mechanisms. Conclusion: NDNF functions as a novel modulator that stimulates revascularization processes. Significance: NDNF represents a novel therapeutic target for ischemic vascular diseas… Show more

“…Recently, NDNF has been found to promote vascularization since expressed at high levels by EC of the ischemic muscle in mice. Intramuscular NDNF overexpression increases perfusion and capillary formation in a murine model of ischemia, while NDNF knock-out resulted in an overall reduction of the EC network (180).…”

“…NDNF was selected as one of potential secreted molecules that are upregulated by ischemia after screening of differentially regulated genes in nonischemic and ischemic muscle tissues of mice. 71 Interestingly, NDNF expression is observed in endothelial cells rather than in myocytes in skeletal muscle tissue. 71 Ischemic insult in the hindlimb leads to enhanced expression in muscle, which is accompanied by increased levels of circulating NDNF.…”
